Cells 'flock' to heal wounds

Like flocks of birds, cells coordinate their motions as they race to cover and ultimately heal wounds to the skin. How that happens is a little less of a mystery today.

Researchers once thought only the cells at the edge of a growing patch of wounded skin were actively moving while dividing cells passively filled in the middle. But that's only part of the picture. Rice University physicist Herbert Levine and his colleagues have discovered that the process works much more efficiently if highly activated cells in every part of the patch exert force as they pull their neighbors along.

There's a need to understand how cells cooperate to protect the site of a wound in the hours and days after injury, said Levine, who has introduced the first iteration of a computer model to analyze the two-dimensional physics of epithelial sheets. He hopes it will give new insight into a process with long-term implications not only for healing but also for understanding cancer, a prime motivator in his research since joining Rice under a grant from the Cancer Research and Prevention Institute of Texas.

A paper on the research by Levine, based at Rice University's BioScience Research Collaborative, and colleagues at the University of California at San Diego and in Germany and France appears this week in the Proceedings of the National Academy of Science.

Levine and his colleagues create computer models of processes seen by experimentalists to flesh out the rules that govern biological systems. "Here, we're combining experimental observations from single cells with general notions from the physics literature to create an integrated way of thinking about this multicellular system," he said.

The new models were prompted by a recent Harvard study showing "that even in the middle of a sheet, cells were dynamically creating heads and tails and were actively moving rather than being passively carried along," Levine said. "This data convinced us that we needed a different way to start to think about the problem."

The body marshals an astounding array of forces to heal wounds, Levine said. Many have to do with cell biology, the internal and external signals that tell a cell when to move, when to stop, when to split and when to die. His team's intent was to focus first on the cell's physical interactions with its neighbors and study what happened if all those complicating factors are eliminated from the simulations.

"We try to unravel what is physics and what is biology," Levine said. "We want to know which parts of the phenomenon don't require sophisticated signaling networks."

In the physics approach to cell motility, he said, "the first thing to do is see how far we can get if we assume that all the cells are following the same rules. Then the only thing that's creating the dynamics of the system is that they're interacting with each other. This is the type of problem that physicists have studied before, usually in nonbiological contexts."

In the Harvard experiment, he said, "They had taken a millimeter-sized tissue that was spreading and showed it wasn't just cells on the end that were pulling on the tissue while the others were spectators." But that work didn't explain how cells in the center of the tissue knew the direction of the edge.

Levine's team looked to the skies for inspiration. "Birds look around and decide which way all their neighbors are flying," Levine said. "The idea that they would move as independent birds but also coordinate is where the idea of flocking came from. This way of thinking hadn't been applied to epithelial tissue motility in wound healing."

What cells "see" are their sticky neighbors, which pull and tug them as they move on lamellipodia, thin sheets that serve as "feet" powered by actin filaments that act something like the treads on a tank. The overlapping lamellipodia of adjacent cells influence each other. "The cells have to figure out which way to go based on competing tendencies: their own tendency to push on the ground and propel themselves forward, and the tendency of their neighbors to try to pull them in various directions," Levine said. "Our basic notion is that as time goes on, these tendencies become correlated as the cell 'tries' to accommodate its conflicting inputs."

The Harvard experimentalists saw that loosely packed cells in the middle of a growing colony tend to swirl in a disorganized manner, and the simulations confirmed this. These swirls are analogous to what is seen in other examples of flocking. But when a wound is introduced, the swirls disappear and cells begin to match direction and velocity and pull toward a common goal. The ones on the edge immediately know which way to go, and everyone else learns from their example. Surprisingly, Levine said, "stickier" cells tend to push forward unevenly, with finger-like protrusions at the leading edge, much like what experimentalists often see.

The simulation model has a long way to go, Levine said. "It's rough around the edges. Biologists who read this will immediately say, 'You've left out all sorts of interesting things we know are happening.'

"Yes, there will be experiments for which this approach will not be sufficient," he said. "It will teach us that in those cases, biology has to exert a more specific role in creating the structures and the motion."

Levine hopes to match the models to current work by experimentalists on motility in cells related to the metastatic spread of breast cancer. "We're a long way from saying anything about this problem," he said. "But that's my overall agenda -- to push my research to where it can make contact with the cancer community."

Obama to name foreign policy aide McDonough as chief of staff

WASHINGTON (Reuters) - President Barack Obama on Friday will name veteran foreign policy aide Denis McDonough as his next White House chief of staff, replacing Jack Lew, as part of an overhaul of his senior staff, a White House official said.

Obama will announce the appointment of McDonough, who worked on Obama's 2008 presidential campaign and is now a deputy national security adviser, at 12:10 p.m. EST at the White House, the official said. McDonough had been widely tipped to succeed Lew after his nomination as Treasury secretary.

In a series of high-level staff changes, Obama will also announce that he is moving White House communications director Dan Pfeiffer to the job of senior adviser and replacing Pfeiffer with his deputy, Jennifer Palmieri.

Potential of differentiated iPS cells in cell therapy without immune rejection

Jan. 25, 2013 ? A new study from Boston University School of Medicine (BUSM) shows that tissues derived from induced pluripotent stem (iPS) cells in an experimental model were not rejected when transplanted back into genetically identical recipients. The study, published online in Cell Stem Cell, demonstrates the potential of utilizing iPS cells to develop cell types that could offer treatment for a wide range of conditions, including diabetes, liver and lung diseases, without the barrier of immune rejection.

Ashleigh Boyd, DPhil, and Neil Rodrigues, DPhil, the study's senior authors, are assistant professors of dermatology at BUSM and researchers at the Center for Regenerative Medicine (CReM) at Boston University and Boston Medical Center (BMC). They also are lead investigators at the National Institutes of Health's Center of Biomedical Research Excellence (COBRE) at Roger Williams Medical Center, a clinical and research affiliate of BUSM.

iPS cells can be developed from adult cell types, such as skin or blood, by returning them to a stem cell state using genetic manipulation. iPS cells are capable of maturing (differentiating) into all the specific cell types in the body, making them a powerful tool for biological research and a source of tissues for transplantation based therapies. Given that iPS cells can be made in a patient-specific manner, there should be great potential for them to be transplanted back into the same patient without rejection. Yet a study published in Nature in 2011 demonstrated that iPS cells transplanted in the stem cell state were rejected in genetically identical recipients.

"The Nature study provocatively suggested that tissues derived from patient-specific iPS cells may be immunogenic after transplantation. However, it never directly assessed the immunogenicity of the therapeutically relevant cell types that could be utilized in regenerative medicine and transplantation," said Rodrigues.

The BUSM researchers evaluated this matter by taking adult cells from an experimental model and deriving iPS cells from them. They then differentiated the iPS cells into three cell types: neuronal (nerve); hepatocytes (liver); and endothelial (blood vessel lining) cells. These three cell types represent each of the three germ layers present during embryonic development -- mesoderm, ectoderm and endoderm. Cells from these layers differentiate and ultimately develop into the body's tissue and organ systems. Using experiments to mirror the potential clinical use of patient-specific iPS cells in cell therapy, the team transplanted each of the differentiated cells into a genetically identical experimental model and found no signs of an elevated immune response or indications of rejection.

The study results suggest that using patient-specific iPS cells should overcome issues of immune rejection in transplantation, which will be a significant problem for potential embryonic stem cell-derived therapies. Immune rejection in transplantation is treated clinically by immunosuppressive drugs but they can have serious side-effects, including the risk of developing cancer.

"If the use of immunosuppressive drugs can be avoided, as may be the case for patient-specific iPS cell based therapies, it would be preferable. Our results are very promising and future work should be directed at assessing whether tissues derived from human iPS cells will similarly lack immunogenicity," said Boyd.

Barclays CEO wants to keep large investment bank

HONG KONG/LONDON (Reuters) - Barclays wants to remain a big player in investment banking even though the British bank is cutting costs and jobs in that business, including at least 70 in its Asian division.

The bank has not finalized the number of jobs to go in Asia but it could be as high as 90, according to people familiar with the plans, who declined to be identified because the matter is confidential.

Antony Jenkins, who took over as Barclays chief executive at the end of August, is expected to cut about 2,000 investment banking jobs globally in a review of the bank's business due on February 12. The bank told UK staff of possible cuts earlier this week.

"Barclays will continue to be a universal bank and it will continue to have a very, very significant and large investment bank as part of that universal banking model," Jenkins told CNBC in an interview from Davos on Thursday.

He declined to specify the scale of cuts in Asia, but said: "We still see opportunities for us in Asia in investment banking and equities, but we also see opportunities for us in other parts of the world so it's a relative, rather than an absolute statement.

"We live in a world where capital is scarce and we have to deploy it against our best opportunities."

Banks globally are having to cut costs to cope with this more hostile climate, partly the result of tougher regulation on the industry after the financial crisis.

The CEO said more regulation and nationalism and a tough economic backdrop would limit revenue growth across the industry and was forcing banks to shrink, pick areas of strength and focus far more on cost-cutting.

The future shape and size of Barclays' investment bank is considered the most critical part of Jenkins' review as the business contributes more than half of group profits.

Jenkins took over from Bob Diamond, who left the bank after a scandal over interest rate rigging for which Barclays was fined $450 million.

The CEO is also attempting to drive through a change in culture and standards after a string of scandals, including Barclays' fine for manipulating Libor interest rates, and mis-selling problems in Britain.

"The industry and Barclays got it wrong on occasions. We were too aggressive, we were too short-term focused and we were too self-serving," he said.

Jenkins said he was confident in his bank's capital strength, after warnings from the Bank of England that UK banks may need to raise capital.

"We are very comfortable with our capital position through the course of this year. We have ongoing discussions with our regulators so we're comfortable with our capital position."

ASIA PULLBACK

In Asia, Barclays had spent heavily to hire a team of veteran investment bankers after the 2008 financial crisis, with the hope of creating a business in the region that would put the London-listed bank in the same league as Goldman Sachs and Morgan Stanley .

But the Asian operation was hit by delays in establishing its equity business, stiffer than expected competition from local firms, and a quick rebound in Asian markets that allowed rivals to regain momentum.

A spokesman for Barclays declined to comment on the scale of cuts there.

Barclays has about 3,000-4,000 investment banking staff in Asia, out of 23,300 globally.

Barclays ranked 8th in investment banking fees earned globally last year, but was only 20th in Asia, according to Thomson Reuters data.

Jenkins said his review - dubbed "Project Transform" - will have repercussions for the bank's corporate structure.

A professor friend of mine recently asked his freshman writing class what makes civilization possible. The students puzzled for a minute and then someone said, ?Cities.? Of course, that?s really just the definition of civilization. ?But what makes those cities possible?? the professor asked. No one could really come up with an answer.

Here were students drawn in many cases from rural areas, some of whom lived on farms; and yet, the most basic energy flow in modern civilization?in any civilization?from farm to city in the form of surplus food was completely opaque to them. My friend remarked to me that a century and half of cheap energy in the form of fossil fuels has attenuated our awareness of energy flows so much that we as a society have become essentially unconscious of energy.

That is a state of mind that could only be the product of energy abundance, of an exceptional period in human history when the surplus energy available to society was so great that the average person simply did not have to think about it. And, so as energy scarcity returns to civilization as the norm, we are being forced?often painfully?to become conscious once again of the energy flows in our daily life. As a whole, human societies are only just beginning to wake up to this new era?except, of course, where life has remained close to the land, and failure to understand and create the necessary energy flows (particularly food) has always been tantamount to a death sentence.

The era of abundance which is now coming to an end created a dependence on a technical-corporate-financial energy elite which took over the provisioning of energy for society as that enterprise became ever more complex?advancing light-years beyond the pick ax of the 19th century coal miner. So long as energy remained abundant, the legitimacy and autonomy of this energy elite went largely unchallenged. Even in countries where the government controls energy resource exploitation, a small elite remains in charge of the process, a process thought to be largely technical in nature.

But the return of energy scarcity in the last decade?in the form of record high oil prices, high and volatile coal prices, wildly volatile natural gas prices in the United States and high prices for liquefied natural gas internationally?all this now threatens to undermine the legitimacy and autonomy of the energy elite. People are beginning to wonder whether it is wise to leave such matters to the putative experts. Should the government take a more active role in directing energy policy? Should communities and households seek to provide their own energy in the form of renewables or even community-owned utilities? Should steps be taken to reduce energy use dramatically to cushion a possible future decline in the availability of energy?

So rattled by these stirrings is the oil and gas industry that it has launched a campaign to convince the public that these hydrocarbons will return to their former abundant state for the long term. This campaign goes by the moniker ?energy independence? in the United States. Of course, energy independence for the United States is unlikely unless it is based on radical reductions in energy use and widespread deployment of alternative energy. Naturally, the oil and gas industry does not have this kind of energy independence in mind.

The industry campaign relies on the clever and deceptive move to redefine what oil is. ?Oil? is now supposed to include natural gas plant liquids (which as you might guess come from natural gas wells and include propane, butane, ethane and pentane), biofuels which include ethanol and biodiesel, and refinery processing gain (which merely measures the well-known scientific fact that the volume of refined products from crude always exceeds the volume of crude oil input). Back in the land of reality worldwide production of crude oil proper, which is defined as crude oil including lease condensate, has been flat since 2005. But, that doesn?t work well with the abundance narrative. So the industry has persuaded government agencies and especially the media to accept this redefinition without them really understanding it.

The campaign also depends on ignoring inconvenient statistical trends. For example, U.S. coal production has been flat since 1998. But the more interesting news is that the total energy content from that coal has been dropping. We are now exploiting coal that is lower and lower in quality. And, the boom in U.S. natural gas production that was supposed to go on for a century has already stalled. Natural gas production in the United States has been flat for over a year.

While the basis for the claim of renewed abundance and energy independence is not borne out by the evidence (see here, here, here and here), the purpose of the campaign is two-fold: 1) To persuade policymakers and the public to open more public lands to fossil fuel exploration while relaxing environmental regulations and 2) to convince both groups that because energy abundance is returning soon, no changes in the current structure of energy production and distribution need to be made. In other words, the technical-corporate-financial energy elite that currently controls the bulk of the world?s energy supply should remain intact and in charge.

Constraints experienced in oil supplies in the 1970s gave a foretaste of how the public might become more engaged in shaping our energy future. Many alternative energy projects involving solar and wind became the responsibility of the household and the individual business owner. The return of cheap energy in the 1980s and 1990s, however, reinforced the power of the energy elite as public awareness of energy plummeted along with energy prices. Since then, wind and solar energy have been increasingly concentrated in the hands of utilities as the energy elite sought to put these forms of energy under its dominion.

But what if we are, in fact, moving into an era of energy scarcity? And, what if our previous energy abundance was, in fact, not merely a product of our inventive genius, but actually the chance concatenation of political, social, economic, technical and even ancient geologic events?

The pronouncements of the fossil fuel industry and its paid spokespersons on Wall Street and elsewhere are clothed in certainty. Their nicely designed graphs of fossil fuel use go up and up into the future. But what if instead we are not at all certain about future energy supplies? What if our previous abundance has been more a product of good fortune than technical competence? The discovery of fossil fuels was essentially a one-time event in the epoch of humans. And, we are on course to use the bulk of those fuels in just two centuries, fuels which took hundreds of millions of years to deposit in the Earth?s crust. Given the uncertainty, shouldn?t we all be engaged in thinking about and acting on our energy future?

The current technical-corporate-financial energy elite knows only how to manage abundance. Abundance smoothes over questions about equity in energy distribution. And, the very ideology this elite adheres to says that energy constraints can only be temporary. In the long run, economic incentives and technical progress will always elicit new sources of energy in ever greater quantities at affordable prices. In short, the framework from which this elite governs our current energy system does not even include the possibility of long-term constraints in energy supply. This is why this elite may even believe its own propaganda about renewed abundance since its worldview doesn?t allow for any other outcome.

No one knows the future, and so no one can know for certain what our future energy supplies will be. The real question is not how to attain certainty, but how to deal with the uncertainty that has always been a part of our energy future. This is a political question, and therefore one that the energy elite does not wish to see as an object of public discussion. It moves the management of energy supply and distribution away from being merely a technical question into one which invites and necessitates broad public participation. Uncertainty should not be an excuse for inaction; uncertainty is the very place where politics intervenes. Knowing with certainty what your actions will bring means you are merely demonstrating a law of physics. Where certainty reigns, there is no need for political give-and-take since the path is clear.

This is why the industry must insist that it is certain about an abundant fossil-fueled future. As an aside, this same industry insisted for two decades that the science behind climate change was uncertain and therefore that we should not take any action. The industry?s highly successful propaganda efforts have forestalled any effective action on curbing fossil fuel emissions. Now, the industry tells us that it is certain about an issue?fossil fuel supplies?that is plagued with far more uncertainty than the link between the burning of fossil fuels and climate change. Greenhouse gases can be measured by anyone, anywhere on the globe with the proper equipment. The worrisome acidification of the oceans (a result of increasing levels of dissolved carbon dioxide) can be verified by anyone near an ocean with the equivalent of a well-calibrated litmus test.

Stores of hydrocarbons, however, are increasingly found miles under the earth or the seabed. The methods for measuring the amount available to society for its use are imprecise at best. And, those measurements depend on nongeologic factors such as future prices and technology, both of which can only be guessed at. No underground resource can be exploited 100 percent. In fact, oil extraction to date has averaged around 35 to 40 percent of the original oil in place. Unconventional oil resources such as tar sands are likely to yield smaller percentages because of the difficulties involved in the extraction and refining of these resources. In addition, such unconventional energy resources are highly sensitive to price because they are so costly to exploit.

As it turns out, we are far more certain about the science behind climate change than we are about the methods used to calculate future world energy supplies. The degree of certainty about climate change calls out for immediate and drastic action because we understand how large the risks are. The degree of uncertainty over future energy supplies calls out for immediate and drastic action because that uncertainty poses large risks for society.

It?s not necessarily the degree of certainty or uncertainty which ought to drive our public policy on any issue, but rather the level of risk which our current state of knowledge suggests is present. In the case of climate change, the risks are known with more certainty each day that we observe massive melting on Greenland and the poles, intense drought and hurricanes, and dying forests made vulnerable to disease and infestation by a warming climate.

In the case of energy supplies, the risks become more apparent because of what we do NOT know. We do not know whether fusion energy will be mastered any time this century. We do not know whether the new unconventional supplies of oil and natural gas will be able to make up for declines in existing fields over time. We do not know whether society as it is currently structured can withstand the very high energy prices in the long term which appear to be necessary to sustain the development of unconventional energy resources.

As the uncertainties over energy grow and as abundance wanes, the technical-corporate-financial energy elite will be faced with a citizenry that has become increasingly aware of its predicament. Scarcity implies rationing. Either the rationing will be done by price?i.e., the rich get what they need and the rest get what they can afford?or it will be done by political mechanisms. The pain comes in either case, and the big question will be whether that pain can find political expression in our unfolding age of energy scarcity?expression that could mean renewed engagement by citizens in an area central to their happiness, their comfort and perhaps even their survival.

When Colin Kaepernick was in college at Nevada, he was a phenomenal player ? the first player in NCAA Division 1 history to pass for 10,000 yards and run for 4,000 yards in his career. But he wasn?t a Heisman Trophy candidate or a first-round draft pick, in part because many people thought the Nevada ?pistol? offense was a gimmick.

Now that Kaepernick has led the 49ers to the Super Bowl from the pistol, the man who invented the offense, former Nevada coach Chris Ault, is celebrating what he has accomplished ? and predicting big things for the pistol in the NFL going forward.

Ault said on NFL Network this morning that contrary to what some people believe, the pistol isn?t predicated on a quarterback running the ball. In fact, Ault said he believes NFL defenses will be able to take away a lot of the runs that Kaepernick does so well. It?s just that when defenses are focused on preventing Kaepernick from getting to the outside, that leaves a lot open in the middle. Ault said he thought Frank Gore?s two touchdown runs against the Falcons in the NFC Championship Game were set up by the defense having to worry about Kaepernick running in the read-option.

?I think defenses will catch up, but here?s the beauty of the pistol: I know we saw Kaep run for 181 yards against Green Bay ? that?s certainly the read part of the game, it?s great ? but what you saw last week is what I believe our pistol brings to the table: Kaep didn?t run it. He read it and handed it off because Atlanta was taking away Kaep on the outside,? Ault said. ?And those two plays, I believe, that Gore scored on, both of them were read-type plays. The beauty of what we?ve done in the pistol and what I?ve seen the 49ers and the Redskins doing, is, it?s not just the read play itself. It?s also the play-action pass off of it.?

Ault said the pistol formation, in which the quarterback is not under center but is closer to the line of scrimmage than a typical shotgun formation, with a running back directly behind the quarterback, makes it harder for the defense to key on the running back.

?When that back sits behind the quarterback, the linebackers do not have a clear view of what he?s doing,? Ault said. ?You can run downhill power games, counters, gaps and all that from the pistol.?

Anyone who thought the pistol was all about the quarterback running the ball after watching Kaepernick?s record-breaking rushing game against the Packers saw in the 49ers? win over the Falcons that that?s not the case: Kaepernick led the 49ers to a win while running just two times for 23 yards.

?I don?t think the NFL quarterbacks are all going to start running the ball 15 times a game,? Ault said. ?But if you?ve got the read in your offense, it is a threat, it?s something you?ve got to be concerned about, and of course if you?ve got a guy like Kaep who can run like a gazelle, you?ve got to be more than concerned with it. You?ve got to put one and a half people on him.?

Ault said that any NFL team, even those with slow, pocket passers, could run its offense out of the pistol.

?They could run the pistol formation,? Ault said. ?They don?t need to run the read part of it. When we first put the pistol in, in 2005 and 2006, that?s all we ran ? we ran the power, the gap, the counters, the zones, the outside stuff. We did not run the read at that time. So the pistol offense, the most important thing there is you can run any offense you?ve been running.?

Ault isn?t suggesting that his offense is the best for every team ? he acknowledged that it has weaknesses and strengths like any other system. But he says it?s not a gimmick. It?s here to stay.

Reports of unrest in small African nation Eritrea

JOHANNESBURG (AP) ? More than 100 dissident soldiers stormed the Ministry of Information in the small East African nation of Eritrea on Monday and read a statement on state TV saying the country's 1997 constitution would be put into force, two Eritrea experts said.

The soldiers held all of the ministry workers ? including the daughter of the president ? in a single room, said Leonard Vincent, author of the book "The Eritreans" and co-founder of a Paris-based Eritrean radio station. The soldiers' broadcast on state TV said the country's 1997 constitution would be reinstated and all political prisoners freed, but the broadcast was cut off after only two sentences were read and the signal has been off air the rest of the day, Vincent said.

By late afternoon there were indications the soldiers' attempt would fail. A military tank sat in front of the Ministry of Information but the streets of the capital, Asmara, were quiet, and no shots had been fired, said a Western diplomat in Eritrea who wasn't authorized to be identified by name.

Vincent stopped short of calling it a coup d'etat and said it wasn't immediately clear if the action was a well-organized coup attempt or what he called a "kamikaze crash."

Later Monday government soldiers surrounded the ministry, an indication the action by the dissident soldiers had failed, said Martin Plaut, a fellow at the Institute of Commonwealth Studies in Britain.

"It looks like it's an isolated attempt by some soldiers who are completely frustrated by what is going on. But it wasn't done in a coordinated manner," Plaut said. "They did seize the television station, they did manage to put this broadcast out, but the government is still functioning calmly. There is nothing on the streets."

Eritrea is an oppressive and politically isolated neighbor of Ethiopia and Sudan situated on the Red Sea that broke off from Ethiopia in the 1990s. The U.S. government's relations with Eritrea became strained in 2001 as a result of a government crackdown against political dissidents, the closing of the independent press and limits on civil liberties, conditions that the State Department says have "persisted to this day."

Isaias Afworki has ruled the country as president and head of the military since 1993.

If the power grab attempt by the dissident soldiers fails, they are likely in for severe punishments, Vincent and Plaut said.

"People call it the North Korea of Africa and that is accurate, so you either win or you're dead, and I think these people are dead," Plaut said. "One can't be absolutely sure but that's what it looks like."

Mission 15 is very short and none of the enemies have to be fought. Cutscenes don?t influence your total mission time. It doesn?t matter if you watch or skip them. All you got to do in this mission is to clear the path for a car. When you encounter enemies just run past them and concentrate on the main objective. Make sure that you never fall down, it will cause you damage and you would have to restart the last checkpoint.

18. Every hero has a weakness (10G) ? You completed Furnace of Souls without taking damage from the furnace

Location: Mission 17: Furnace of Souls
During the entire 17th mission you may not be damaged by the fire that is emitted from the ground every 7 seconds. The fire can only damage you while you are in the air, so do your best to move between platforms very quickly. While standing on solid ground you are save. Restart the last checkpoint immediately if you fall down or are damaged by the fire. During the fight sequences make sure that you are not pushed into the fire by your enemies. The trophy / achievement will unlock after the mission is over.

19. It?s only the rain (10G) ? You Killed 10 enemies by pushing them into the Hurricane ride

Location: Mission 1: Found
After walking past a lost soul you will come to an area with a big hurricane ride (some sort of carousel that is spinning in the air). The screen will say ?Death? and 10 enemies will spawn. Lure the enemies below the hurricane ride and press Circle (PS3) / B (X360) to launch them into the air. They will die instantly if the hurricane ride hits them. Do this to all 10 enemies.

20. A man with guts and honor (10G) ? You reached the end of the descent on Mission 6 having killed all of the enemies

Location: Mission 6: Secret Ingredient
Immediately after the mission starts you will have to reach the end of the descent within 2 minutes. It?s recommended to attempt this trophy / achievement on human (easy) difficulty because enemies will die after just a few hits. There are a total of 10 enemies. Make sure to kill all of them and open the door at the end of the descent within the 2 minute limit.

21. Now my coat?s all charred (10G) ? You navigated the Sky Bridge without hitting the lasers

Location: Mission 16: The Plan
Towards the end of the 16th mission you will come to a corridor with several laser barriers. To get past them you need to vary your jump height and use your floating skill (L2+X on PS3 / LT+A on X360). For one barrier you must use the floating skill from the ground, without jumping. If you touch a barrier you can still restart the checkpoint and try it again.

22. Where does the time go? (10G) ? You completed a level in 2 minutes or less

Location: Mission 15: The Trade
Mission 15 is very short and none of the enemies have to be fought. Cutscenes don?t influence your total mission time. It doesn?t matter if you watch or skip them. All you got to do in this mission is to clear the path for a car. When you encounter enemies just run past them and concentrate on the main objective. Make sure that you never fall down, it will cause you damage and you would have to restart the last checkpoint.

23. For Tony Redgrave (10G) ? You killed 50 enemies using nothing but firearms

24. In the name of my father (10G) ? You killed 100 enemies using nothing but Demon weapons

25. You?ll never have her fire (10G) ? You killed 100 enemies using nothing but Angel weapons

From the Main Menu, select Secret Missions, and start Angelic Warfare. This will start a mission where you have to kill all enemies within the time limit; where only Angel weapons cause damage to the enemies.

26. Impressive (10G) ? You have slayed 100 Demons

27. Bring it on! (20G) ? You have slayed 1,000 Demons

28. Looks like we have a winner (30G) ? You have slayed 5,000 Demons

29. Sensational! (10G) ? You gained a SSS Style Rank during combat

Location: Mission 11: The Order
Make sure to set your difficulty to human (easy) difficulty and start mission 11. In the very beginning you will have to fight several Stygians. Start with an Arbiter Tremor (R2+Circle on PS3 / RT+B on X360), followed by an Osiris Karma attack (L2+Triangle on PS3 / LT+Y on X360). Switch to the Aquila for a few strikes and you should already have the SSS bonus. Try to hit as many enemies as possible with every attack and avoid repeating the same move.
Restart the mission if all enemies die before you get the SSS Style Rank.

Facebook Graph Search Doesn't Lend Much Help to Advertisers or ...

Facebook today announced a new product called Facebook Graph Search, now in beta.

Unlike Google and other web search engines, Facebook Graph Search will allow users to find Facebook-only content in four main areas (example queries from the Facebook press release below):

People: ?friends who live in my city,? ?people from my hometown who like hiking,? ?friends of friends who have been to Yosemite National Park,? ?software engineers who live in San Francisco and like skiing," "people who like things I like," "people who like tennis and live nearby"

Photos: ?photos I like,? ?photos of my family,? ?photos of my friends before 1999,? "photos of my friends taken in New York," ?photos of the Eiffel Tower?

Places: ?restaurants in San Francisco,? ?cities visited by my family,? "Indian restaurants liked by my friends from India," ?tourist attractions in Italy visited by my friends,? ?restaurants in New York liked by chefs," "countries my friends have visited"

Interests: ?music my friends like,? ?movies liked by people who like movies I like,? "languages my friends speak," ?strategy games played by friends of my friends,? "movies liked by people who are film directors," "books read by CEOs"

This is not a new search engine, it's a more advanced way to browse existing content on Facebook. Facebook already had a search bar on the top but it was limited to names and page titles. With the new search bar users will apparently be able to type actual queries to find the information they are looking for. Google killer? Not likely, but it could keep users on Facebook longer and help them find private info.

Facebook Graph Search appears to be a great way to search through your stuff, but from an online advertiser's perspective, I'm unimpressed. It remains unclear how advertisers will be able to use this Graph Search product to better market and sell their products to Facebook users. Search can be great for ad targeting because it allows marketers to direct ads about their products and services to the right people at the right time. Unfortunately, the new Facebook Graph Search capabilities are limited to people, locations, photos, and other types of searches that are weak in revealing commercial intent. In typical Facebook fashion, the official Facebook Graph Search announcement does not outline any benefit for its advertisers.
